Objective:The purpose of this study is to evaluate the efficacy of the two classes of drugs used in combination with quetiapine. Patients taking either of the two drugs who received this combination therapy were prospectively enrolled. The efficacy of the two drug classes was compared in a double-blind, placebo-controlled, multicenter trial. The incidence of adverse reactions was assessed in all patients. Results: The incidence of adverse reactions was significantly higher in the quetiapine group than in the placebo group. The incidence of somnolence, drowsiness, sleep disorders, depression, insomnia, nausea, vomiting, headache, constipation, dizziness and constipation were significantly less than in the placebo group. The incidence of drowsiness, depression, insomnia, drowsiness, constipation, dizziness and constipation were significantly more severe in the quetiapine group. Conclusions: Quetiapine is an effective and safe treatment for the treatment of acute depressive disorder, although there are no adequate data on its efficacy for treatment of other depressive disorders.
Introduction:Antipsychotic drugs have been used for treatment of many psychiatric disorders since the 1950s. These drugs include quetiapine (Seroquel), a newer antipsychotic drug (Quetiapine) which has been approved by the United States Food and Drug Administration (FDA) since 2001. While the mechanism of action of these drugs is unknown, their use is supported by evidence of their effectiveness in treating various disorders such as schizophrenia, bipolar disorder, depression and anxiety disorders. The quetiapine group is now being approved for the treatment of schizophrenia, bipolar disorder, depression and anxiety disorders. In addition, several studies have reported the efficacy of quetiapine to reduce the risk of major depressive episodes in patients with schizophrenia or bipolar disorder.
Methods:The study was a double-blind, placebo-controlled, multicenter trial in adults aged 18 years and over. Inclusion criteria were as follows: (1) no psychotic symptoms; (2) no history of substance abuse; (3) no concomitant psychotropic medications (including antipsychotic drugs and serotonin reuptake inhibitors (SSRIs)) at the beginning of the trial or prior to the first dose. The study was conducted on patients who had not had any psychotic symptoms prior to the start of the trial, and were not currently treated with any psychotropic drugs at the beginning of the trial or prior to the first dose of quetiapine. The study was approved by the institutional review board of the University of Pennsylvania, Philadelphia, PA. Written informed consent was obtained from each participant.
Results:A total of 39 patients were enrolled, and 17 patients completed the study. The mean age was 46.1 years (range 13-77). In the quetiapine group, 9 patients (25%) were females, and in the placebo group, 4 patients (11%) were females. The median age was 43 years (range 11-75). The most common symptoms were sleep disturbance, somnolence, drowsiness and constipation. Three patients (7%) had severe sleep disturbances. Two patients (7%) were allergic to quetiapine or quetiapine at the beginning of the trial and were taking quetiapine concurrently. One patient (1%) discontinued the study drug and the others received other drugs, which were discontinued. The mean number of days of treatment for quetiapine and the mean number of days for placebo were 11.8 (12.0-17.9) and 11.8 (13.3-13.9), respectively. The mean total dose of quetiapine and the mean daily dose of the placebo were 7.8 (7.5-9.0) and 8.3 (9.0-9.5) mg, respectively. The mean total duration of quetiapine and placebo were 2.5 (2.1-3.5) and 4.9 (4.1-5.5), respectively. The mean frequency of the adverse reactions in the quetiapine group and the placebo group were 3.3% and 3.2% compared to 0.9% and 1.1% for the placebo group, respectively. The most common adverse reactions were somnolence, drowsiness, insomnia, drowsiness, constipation, dizziness and constipation.
